FBI Director Kash Patel said the agency was "closely monitoring" the shooting.
"Agents will be on the scene quickly and the FBI stands in full support of the ongoing response and investigation," he posted on X.
Video circulating on social media showed Kirk sitting on an outdoor stage speaking to a large crowd at Utah Valley University in Orem when a shot was fired. Kirk recoiled as blood gushed from his neck, and the audience fled in panic.
A suspect was taken into custody by campus police, Utah Valley University said on X, adding that Kirk was removed from the scene by his security.
His condition is unknown.
"We must all pray for Charlie Kirk, who has been shot," Trump wrote on his Truth Social account. "A great guy from top to bottom. GOD BLESS HIM!"
"Say a prayer for Charlie Kirk, a genuinely good guy and a young father," said Vice President JD Vance on X.
The 31-year-old does not hold elected office but is a powerful force in grassroots conservative politics, leading youth organization Turning Point USA, hosting a popular podcast and attracting millions of followers on social media.
He spoke at Trump campaign rallies in last year's election and addressed the Republican National Convention.
Political violence is a growing concern in the deeply polarized country, affecting both Republicans and Democrats at all levels of government.
Trump himself was the victim of an assassination attempt during the campaign in which he was slightly injured at a rally in Pennsylvania.
